Polyethyleneglycol (PEG) is the most widely used polymer in the formulation of liposomes. The PEG groups are generally conjugated to the ethanolamine moiety of phosphatidylethanolamine (PE), such as DSPE-PEG2000-X. The single hydroxyl group of PEG lipid can be coupled with small molecule drugs, antibodies, and groups for click chemistry and thus formulates different functional liposomes. However, another kind of methoxy PEG is closed at one end and cannot be functionalized, which is commonly used in stealth liposomes. When injected into the body, liposomes with hydrophilic PEG chains will form brush-like barriers, which can reduce liposome aggregation and liposome-protein interactions and therefore increase the blood clearance time.
